A structured PDMS stamp functionalized with streptavidin is deposited on a Surf (Whitesides method). Through hydrophilic transfer, the streptavidin gets absorbed on the surf. Without any labeling, a Sarfus optical observation allows imaging directly the pattern (grid of 4 µm*4µm) of streptavidin and its defects, which is 4nm-thick.
